FAQ: What is the smallest volume of elution buffer that can be used with the Monarch® Plasmid Miniprep Kit (NEB #T1010)?
The protocol is optimized to provide good yields with elution volumes as low as 30 μl of elution buffer. Lower volumes can be used to increase the concentration of the eluted DNA, but the overall yield will be reduced due to incomplete wetting of the membrane. Elution volumes higher than recommended (>100 μl) can also be used, but the eluted DNA will have a lower concentration.
