Developing computational tools to enable synthetic biology research

As part of the long-term collaboration with the Lohman lab, we develop and maintain NEBridge® Ligase Fidelity Tools that enable synthetic biology research and power high-complexity and high-fidelity Golden Gate Assemblies (ligase fidelity data and tools).

Ligase Fidelity Tools enable examining and optimizing overhangs for Golden Gate Assembly, and oligo-based gene synthesis.

In collaboration with Sean Lund and other NEB Scientists, we take further these technologies to enable highly parallelized construction of DNA from low-cost oligonucleotide mixtures (https://pubs.acs.org/doi/10.1021/acssynbio.3c00694).

The image shows nucleotide sequences turned into barcoded fragments, which are then combined into an oligo pool.
